자가면역질환 연구실

Laboratory of Autoimmunity

본 연구실은 류마티스 내과 연구실에서 2004년 이은영 교수에 의해 구성되었으며, 다양한 자가면역질환 환자 유래 검체의 임상 정보기반 분석을 통해 병인 기전과 치료 타겟 탐색을 목표로 연구를 이끌어가고 있습니다. 그동안 다수의 석사, 박사를 배출하는 성과를 내었고, 현재 박사급 연구원 1명, 석사급 연구원 2명, 박사과정생 1명, 석사 과정생 1명으로 구성된 우리 팀은 환자 유래 세포 및 자가면역질환 동물 모델을 이용하여 질환의 기전 규명 및 최적화된 치료 전략을 위한 연구를 진행 중입니다. 2014년부터 연구중심병원 염증유닛의 3세부를 맡아 진행하고 있습니다.

연구내용

악성종양에서의 면역반응과 자가면역질환에서의 면역반응은 서로 공유 혹은 상반되는 면들을 가지고 있습니다. 따라서 이러한 질환들에서 진행되는 반응들을 서로 비교, 연구함으로써 공통되거나 혹은 질환 특이적인 타깃을 찾는 노력이 필요합니다. 대표적으로 악성 종양을 동반하는 자가면역 질환인 염증성 근육염 환자 근육조직의 대사산물을 분석하여 기능이 저하된 미토콘드리아를 목표로 하는 연구와 개발 플랫폼을 활용한 PCT 국제 특허 출원 (Patent Cooperation Treaty, PCT) 및 임상 시험 계획 구축을 진행하고 있습니다. 다기관 류마티스관절염/간질성 폐 질환 환자 코호트에서 수집한 검체를 분석하여 혈액 및 영상 바이오 마커를 발굴하며, 관절염/폐질환 동물모델을 구축하여 질환 진행 및 치료에 따른 폐병변의 변화와 단일세포전사체 분석을 진행하고 있습니다. 정밀 치료를 위한 질환 기전 규명 및 신규 타깃 디스커버리를 목표로 강직척추염 환자에서 약제 치료 전후 임상자료 코호트 확립 및 종적 시료 수집을 진행하고, 단일세포 전사체 분석 등을 활용한 연구를 진행 중 입니다.

협업은 의료와 의학을 함께 발전시키는 데 매우 중요합니다. 본 연구팀은 서울대학교병원내 임상의학 연구진들 이외에도 서울의대 해부학교실, 생화학교실, 카이스트 면역학, 유전체연구팀을 비롯하여 지놈인사이트, 와이바이오로직스, 이뮤노포지, 파이안바이오 등의 국내 바이오벤처와 개방적인 협력연구를 진행하고 있습니다. 미국 NIH 희귀질환연구진, 코넬의대 면역학/골면역학 연구진들과 협력연구를 통해 논문을 출간한 바 있습니다.

연구목표

신규 치료 타겟 및 바이오마커를 발굴하고 실제 치료에 적용하기 위해 염증성 근염 (피부근염/다발성근염) 근육조직과 혈청 샘플 에서 대사체 프로파일링 분석을 계획하고 있습니다. 또한, 단일 세포 유전체 분석과 후성유전체 연구를 융합하여 특정 질환 및 생물학적 제재 치료 시 면역세포 간의 다양한 상호작용과 변화를 밝히고자 합니다. 연구 결과를 바탕으로 병인 기전에 관여하는 주된 세포 구성을 밝혀내고, 이를 선제적 치료에 적용하고자 합니다.
