Real time quantitative PCR 이용 안내

암연구소에 설치되어 있는 real time quantitative PCR (실시간 유전자 증폭 정량 측정 장치) 이용에 대하여 안내해 드립니다. 보유하고 있는 real-Time PCR (실시간 유전자 증폭 정량 측정 장치) 자세한 재원은 아래와 같으며, 기기담당 연구원이 분석을 도와드립니다. (전화 3668-7468).

Real-time PCR를 이용하실 분은 암연구소 홈페이지 (http://cri.snu.ac.kr)의 공용기기예약에서 예약하시고 사용설명서를 작성해서 담당자 이 메일로 보내주시면 됩니다.

예약날짜에 서울대학교 암연구소 9층 911호에 sample(reference 포함), primer또는 probe(target, endogenous control)을 가져오신 후 담당자에게 신청하시면 됩니다.

Taqman probe의 endogenous control (Human)은 준비되어 있습니다.

< 의뢰 시 체크사항 >

   ① Primer design

      - Tm 58℃-60℃

      - No more than 2 G+C's in last 5 bases on the 3' end

      - Amplicon length maximum = 150bp

   ② Probe design

      - Tm 10℃ higher than primers (68℃-70℃)

      - 5' base may not be G

      - Select probe with more Cs than Gs

      - Synthesize probe with a Reporter dye at the 5' end and Quencher Dye at the 3'end

   ③ cDNA : 0.5-1.5㎍/㎕
